Фосфолипидная система с адресным пептидом ангиопеп-2 для доставки хлорина е6 в исследовании in vitro

Аннотация
Полученная ранее фосфолипидная наносистема для доставки фотосенсибилизатора хлорина е6 была модифицирована нацеливающим лигандом – олигопептидом ангиопеп-2, обладающим высокой способностью к трансцитозу через гематоэнцефалический барьер. Данное свойство ангиопеп-2 особенно актуально для направленной доставки терапевтических и диагностических агентов в патологические (опухолевые) очаги мозга. Согласно проведенному анализу физико-химических параметров разработанной композиции, размер частиц составил 31.98±1.98 нм (PdI 0.453±0.03), значение ζ-потенциала соответствовало -27.43±1.14 мВт; при этом субстанция практически полностью (98.6±0.43%) была встроена в наночастицы. В эксперименте in vitro было показано увеличение общего накопления и интернализации хлорина е6 на клеточной линии глиобластомы человека U-87 MG в варианте с содержащей адресный пептид фосфолипидной формой по сравнению со свободной субстанцией на 33% и 40% соответственно. Исследование цитотоксического действия без облучения не выявило различий между образцами в диапазоне концентраций от 0.125 мкг/мл до 2.5 мкг/мл (по хлорину е6). Изучение фотоиндуцированной активности (доза облучения 1.5 Дж/см2) показало, что значение IC50 для полученной композиции было ниже в 1.33 раза по сравнению со свободной субстанцией и составило 2.85±0.1 мкг/мл. Результаты проведенных экспериментов свидетельствуют о перспективности разработанной композиции и целесообразности её дальнейшего исследования in vitro и in vivo.
