Аннотация

Онкологические заболевания представляют серьезную проблему для общественного здравоохранения, унося миллионы жизней ежегодно. Современные методы лечения онкологий все еще имеют существенные ограничения, такие как побочные эффекты, ограниченная эффективность при метастазировании и рецидивах, и высокая стоимость, поэтому разработка новых подходов к онкотерапии является важной и приоритетной задачей. Бактерии можно рассматривать как универсальный и пластичный природный биоматериал, который обладает свойством самостоятельно передвигаться и проникать в труднодоступные участки человеческого тела, доставляя терапевтические агенты непосредственно к раковым клеткам. Также бактерии обладают естественной иммуногенностью, что позволяет привлечь иммунные клетки в зону опухоли и повысить эффективность иммунного ответа. Неудивительно, что фантастическая на первый взгляд идея создания бактериобота – автономного микроробота на основе живой бактерии для доставки терапевтического груза – становится все более популярной. В настоящем обзоре рассмотрены основные принципы конструирования бактериальных микророботов, способы их нацеливания на мишень и снабжения терапевтическим грузом, также затронуты вопросы безопасности бактериоботов для человека и перспективы их применения в лечении злокачественных новообразований.
